My Parents live in Kissimmee so I go down about 2 times a year. The list is way too long for things that need to be seen. Heh Add to that there are many new things that are about to open.
Short list of my "must sees"
MGM... Star Tours (going to go through a renovation and program change)
Star Wars Jedi Academy. Fun show.
Muppets 3D...take some time to walk around the building and take special note to all the signs and extras.
Animation studio walk through. Pretty neat
Block Party Parade....high energy fun
Tower of Terror
Rockin Roller Coaster
Animal Kingdom Finding Nemo musical show
Expedition Everest! Fun roller coaster
Safari bus tour
It's Tough To Be A Bug
Disney World
Monsters, Inc Laugh Floor
Haunted Mansion and Pirates have had upgrades but are still wonderful entertainment staples.
Everything else
EPCOT
Eat at Morocco! Good food everywhere
It will also be their Food and Wine Festival in October. (also "Eat to the Beat" so look for performer schedules closer to date. It's where I saw Big Bad Voodoo Daddy.)

The Sea with Nemo and friends.
TEST TRACK
Mission Space (it's a centrifuge ride...I believe they now have a stationary version for people with issues)
Pretty much anything there is fun.
